Whole cell lysates (15 µg total protein) from indicated cell lines were resolved by gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-SNAIL (clone W22252A). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control.     MCF7 cells (low expressing cell line, panel A) and BeWo cells (high expressing cell line, panel B) were fixed with Fixation Buffer (Cat. No. 420801), permeabilized with 0.5% Triton X-100, and blocked with 5% FBS. Cells were then stained with Purified anti-SNAIL (clone W2222A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm.

Description

SNAIL is a zinc finger transcriptional repressor that is involved in cell movement, adhesion, and survival. SNAIL plays a pivotal role in the epithelial to mesenchymal transition (EMT) during embryonic development. SNAIL represses the expression of E-cadherin in epithelial cells leading to the disassembly of cell-cell junctions, loss of cell polarity, and increased migration. In pathological contexts, SNAIL can promote tumor progression and metastasis by allowing cancer cells to invade surrounding tissues.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use of this antibody in immunocytochemistry (ICC), we recommend fixation/permeabilization with PFA 2% and Triton X-100 0.5%. We do not recommend PFA/methanol or methanol alone.
