Serine/Threonine Kinase 4, also known as STK4 or MST1, is essential for regulating cellular processes like stress response, apoptosis, and organ size control. Functionally, STK4 is a stress-activated kinase that can induce apoptosis by entering the nucleus and causing chromatin condensation and DNA fragmentation. It plays a crucial role in the Hippo signaling pathway, which restricts cell proliferation and promotes apoptosis, thereby preventing tumor growth. STK4 is regulated through its interactions with other proteins in a kinase cascade, including STK3/MST2 and LATS1/2, which further modulate proteins involved in cell growth and apoptosis. Mutations or deficiencies in STK4 can lead to severe immunodeficiencies and other health issues.

-
Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
- Application Notes
-
For use of this antibody on immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend using Tris-EDTA buffer (Cat. No. 422704) for antigen retrieval. We do not recommend citrate buffer.
Antigen Details
- Structure
- STK4 is a 487 amino acid protein with a molecular weight of approximately 55 kDa.
- Distribution
-
The STK4 protein is primarily localized in the cytoplasm but upon activation can be found in the nucleus
- Function
-
The regulation of STK4 involves several mechanisms:
- Phosphorylation: STK4 is activated through phosphorylation
- Protein Interactions: STK4 interacts with various proteins in the Hippo signaling pathway, including STK3/MST2 and LATS1/2
- Stress Response: STK4 is activated in response to cellular stress
- Negative Regulation: STK4 can negatively regulate certain pathways, such as the TBK1-IRF3 signaling pathway
- Interaction
- STK4 interacts with various proteins in the Hippo signaling pathway, including STK3/MST2 and LATS1/2.
