Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-TAZ (WWTR1) (clone W22256A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426318) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining of Purified anti-TAZ (WWTR1) (clone W22256A) on formalin-fixed paraffin-embedded human colon tissue. Following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0), the tissue was incubated either without (panel A) or with (panel B) Purified anti-TAZ (WWTR1) (clone W22256A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 50 μm -
U251-MG cells were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton-X for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were either stained with Purified Rat IgG2a, κ Isotype Control (Cat No. 400502) (panel A) or Purified anti-TAZ (WWTR1) (clone W22256A) (panel B), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). TAZ, also known as WWTR1, is a critical regulator in various cellular processes including cell proliferation, differentiation, and organ development. It is a key component of the Hippo signaling pathway, which controls organ size by regulating cell growth and apoptosis. TAZ functions primarily as a transcriptional co-activator, partnering with transcription factors such as TEAD to promote the expression of genes involved in cell cycle progression and survival. Additionally, it plays a role in maintaining stem cell pluripotency and promoting epithelial-mesenchymal transition (EMT), a process important in development and cancer metastasis. Aberrations in TAZ activity are linked to various cancers, highlighting its significance in both normal physiology and disease pathology. TAZ should not be confused with tafazzin, which was previously also known as Taz/Taz1.

- Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 0.5 - 1.0 µg/mL is suggested. For intracellular flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody (clone W22256A) in immunocytochemistry (ICC) we recommend fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with either 0.5% Triton-X or 100% ice-cold methanol. Permeabilization with 100% ice-cold methanol results in a dimmer signal compared to 0.5% Triton-X. Fixation/permeabilization with 100% ice-cold methanol alone is not recommended.
For use of this antibody (clone W23013A) in intracellular flow cytometry (ICFC), we recommend fixation/permeabilization using fixation buffer (Cat. No. 420801) followed by True-Phos™ Perm Buffer (Cat. No. 425401).
For use of this antibody (clone W23013A) in immunohistochemistry (IHC), we recommend antigen retrieval with Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422703). Antigen retrieval with Sodium Citrate H.I.E.R. (Cat. No. 928502) is not recommended.
