Western blot of purified anti-VPS35 antibody (clone A17084A). Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain lysate; Lane 3: 20 µg of mouse brain lysate; Lane 4: 20 µg of rat brain lysate; Lane 5: 20 µg of HEK293 lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

Description

VPS35 is the largest subunit of the retromer complex, which plays an important role in retrograde protein transport from the endosome to the trans-Golgi network. VPS35 binds to VPS26 (VPS26A or VPS26B) and VPS29 to form the vacuolar protein sorting subcomplex (VPS), which in turn interacts with different nexins to form different retromer variants that transport different targets. D620N mutation in VPS35 has been implicated as a causative genetic factor for developing late-onset, autosomal dominant familial Parkinson’s disease, and could serve as potential therapeutic target for PD.
