Description

Fc gamma RIII, also known as CD16 is a low affinity IgG Fc receptor with a broad range of different isotypes. In mouse, it is encoded by a single gene, whereas in human, there are two distinct forms (Fc gamma RIIIA/ CD16a and Fc gamma RIIIB/ CD16b), encoded by two nearly identical genes which are expressed in a tissue-specific manner. Fc gamma RIIIA is a 50-65 kD type I transmembrane protein. It is expressed on the surface of NK cells, activated monocytes, macrophages, and placental trophoblasts in humans. Fc gamma RIIIB is a 48 kD GPI-anchored protein. Its extracellular domain is over 95% homologous to that of Fc gamma RIIIA, and it is expressed specifically on neutrophils, eosinophils and a subset of basophils. Several alleles of Fc gamma RIIIB, such as NA1, NA2 and SH, have been identified on PMNs. The alloantigens of the neutrophils are involved in alloimmune neonatal neutropenia. Fc gamma RIIIB binds to aggregated IgG or IgG-antigen complex, but not monomeric IgG. Human Fc gamma RIIIB binds to different human IgG isotypes with distinct affinities; it binds to IgG1 and IgG3, but not IgG2 and IgG4. Glycosylation of Fc gamma RIIIB regulates its affinity to ligand IgG. Ligation of Fc gamma RIIIB results in increased cytosolic calcium, lysozyme release, and the recruitment of neutrophils to immune complexes. It has been reported that Fc gamma RIIIB is cleaved by ADAM17 and released from plasma membrane of the activated neutrophils during apoptosis. Soluble form of Fc gamma RIIIB can inhibit B cell proliferation and immunoglobulin production.

Molecular Mass
The 199 amino acid recombinant protein has a predicted molecular mass of approximately 22.7 kD. The DTT-reduced and non-reduced protein migrates at approximately 45-55 kD by SDS-PAGE. The predicted N-terminal amino acid is Thr.

> 95%, as determined by Coomassie stained SDS-PAGE.

Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.

Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.

Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
