Frizzled-8 (FZD8), a G-protein coupled receptor with seven transmembrane domains (7TM), is one member of ten Frizzled protein family. Frizzled proteins are the principal receptors for the Wnt family of ligands, mediating canonical Wnt signaling. Frizzled proteins are present in most of the animals, but not present in plants or in simpler (single cell) eukaryotes. In the presence of Wnt ligand, Wnt binds to extracellular cysteine rich domain (CRD) of Frizzled. A receptor complex forms between Frizzled and low-density lipoprotein receptor-related protein 5/6 (LRP5/6). Subsequently, Dishevelled (Dvl) recruitment by Frizzled leads to LRP5/6 phosphorylation and recruitment of scaffolding protein, Axin. This disrupts Axin-mediated phosphorylation/degradation of β-catenin, allowing β-catenin to accumulate in the nucleus where it serves as a coactivator for TCF to activate Wnt-responsive genes. Human FZD-8 contains 694 amino acids (aa) with a 248 aa extracellular domain (ECD) (aa 28-275) that includes a highly conserved cysteine-rich region (CRD), the 7TM region (aa 276-605), and a C-terminal cytoplasmic domain (aa 606-694). According the identity of aa sequences, frizzled proteins can be divided into four subgroups: subfamily 1 (FZD1, FZD2, and FZD7), subfamily 2 (FZD5 and FZD8), subfamily 3 (FZD9 and FZD10), subfamily 4 (FZD4) and subfamily 5 (FZD3 and FZD 6). FZD8 shares about 70% identity with other frizzled proteins. hFZD-8 is abundant expressed in fetal kidney and also expressed in fetal brain, fetal lung and adult kidney, adult heart, adult pancreas and adult skeletal muscle. Knockout of FZD8 in mice has no apparent phenotype, but leads to reduced ureteric bud growth and a reduction in kidney size. It has been reported that FZD8 can promote the cell proliferation and metastasis of renal cell carcinoma. Targeting Human FZD-8 suppresses the migration and invasion of breast cancer and non-small cell lung cancer (NSCLC) cells through inhibiting Wnt/β-catenin signaling pathway. FZD-8 also participates in non β-catenin signaling pathway. FZD8 integrates Wnt-11 and TGF-β signals to promote epithelial-to-mesenchymal transition (EMT) independently of β-catenin. FZD-8 can serve as a potential therapeutic target in many human cancers.

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

- ED50 = 80 – 420 ng/mL as measured by its ability to inhibit Topflash reporter activity in HEK293T human embryonic kidney cells induced by 50 ng/mL of recombinant mouse Wnt3a (Cat. No. 772301).

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
