Seven IGFBPs have been described to modulate the IGF activity. IGFBPs transport IGFs, in which they may inhibit mitogenesis, differentiation, survival, and other IGF-stimulated events. IGFBPs structurally are characterized by three domains: the amino-terminal, the carboxi-terminal, and an intermedia or variable L-domain. Members of the IGFBP family exhibit 67–70% structural homology. The greatest homology among the IGFBPs is in the N- and C-terminal regions. Some IGFBPs bind to the extracellular matrix (IGFBP-2, -3, -5, and -6). In fact, a heparin-binding domain (HBD) has been identified in the C-terminal region of these binding proteins. IGFBP-5 is the most conserved IGFBP in mammals. Additionally, human and mouse IGFBP-5 are 97% identical. The IGFBPs bind IGFs with high affinity (kD ∼0.1 nM), and this binding restricts the interaction of IGFs to their receptors. The release of the IGFs from IGFBP-5 is mediated by proteolysis; it has been described that IGFBP-5 is cleaved by ADAM-12, ADAM-9, C1s, and PAPPs (pregnancy-associated plasma proteases) A and A2. IGFBP-5 plays a key role in tissue repair and regeneration. IGFBP-5 is overexpressed in fibrotic lung tissues from patients with idiopathic pulmonary fibrosis. In vitro, IGFBP-5 induces the production of ECM components while in vivo, IGFBP-5 initiates the development of dermal and pulmonary fibrosis. In addition, IGFBP-5 induces lung fibroblast and mononuclear cell migration in an IGF-I–independent manner. IGFBP-5 plays an important role in mammary development, involution, and breast cancer metastasis. IGFBP-5 has nuclear localization sequence that mediates β-importin-dependent translocation to the nucleus, where it interacts with the vitamin D receptor and modulates the vitamin D response in osteoblasts.

- Molecular Mass
- The 252 amino acid recombinant protein has a predicted molecular mass of approximately 28.57 kD. The DTT-reduced and non-reduced protein migrates at approximately 31-35 kD by SDS-PAGE. The predicted N-terminal amino acid is Leu.

- >95%, as determined by Coomassie stained SDS-PAGE.
- Formulation
- 0.22 µm filtered protein solution is in 40mM sodium citrate, pH4.
- Endotoxin Level
- Less than 0.1 EU per µg cytokine as determined by the LAL method.
- Concentration
- 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- When recombinant human IGFBP-5 is immobilized at 0.5 μg/mL, biotinylated recombinant human IGF-II binds in a dose-dependent manner with ED50 range of 0.9 – 5.4 ng/mL. HRP Avidin (Cat. No. 405103) was used to detect the binding.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
- Does specific activity of a recombinant protein vary between lots?

Specific activity will vary for each lot and for the type of experiment that is done to validate it, but all passed lots will have activity within the established ED50 range for the product and we guarantee that our products will have lot-to-lot consistency.
