Western blot of purified anti-ALDH2 antibody (clone 8D4-D6-D11). Lane 1: Molecular weight marker; Lane 2: 10 µg of human brain lysate; Lane 3: 30 µg of mouse brain lysate; Lane 4: 10 µg of rat brain lysate. The blot was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system. -
IHC staining of purified anti-ALDH2 antibody (clone 8D4-D6-D11) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 5 µg/ml of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale Bar: 50 µm -

ALDH2 is a member of the aldehyde dehydrogenase family. In the human brain, ALDH2 is the major aldehyde metabolizing enzyme that is essential for the metabolism and detoxification of endogenous and exogenous aldehyde substrates. In line with this, recent studies have reported that ALDH2 level is dysregulated in the brains of patients with Parkinson’s and Alzheimer's diseases, suggesting toxic aldehydes as a contributing factor to neurodegenerative diseases and a potential therapeutic target.
