Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-CDK1 (clone A20004C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:25000 dilution (lower). Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker -
ICC staining of purified anti- CDK1 (clone A20004C) on HeLa cells. The cells were fixed and permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 10 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. The image was captured with a 60X objective. -
Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of purified mouse IgG2b, κ isotype control (Cat. No. 400302) or purified anti-CDK1 (clone A20004C). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a mouse control antibody against a separate epitope of CDC2. IHC staining of purified anti-CDK1 (clone A20004C) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated without (panel A) and with (panel B) 5.0 µg/mL of primary antibody followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 10X objective. Scale bar: 50 µm

Cyclin-dependent kinase 1 (CDK1) is a serine/threonine kinase that is ubiquitously expressed in eukaryotes. CDK1 is a catalytic subunit of a protein kinase called the M-phase promoting factor that universally induces entry into mitosis in eukaryotes. CDK1 has been shown to interact with numerous proteins including cyclin A1, cyclin A2, cyclin B1, cyclin B2, BRCA1, dynamin 2, Fyn, Lyn, and p53. In addition to regulation by protein-protein interactions, CDK1 is controlled by phosphorylation, particularly at conserved tyrosine residues. In humans, CDK1 is encoded by the gene CDC2 (cell division control protein homolog 2).

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
