U-251 MG cells (positive cell line, panel A) and HeLa cells (low expressing cell line, panel B) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 1:50 dilution of Purified anti-Nestin (clone Poly22A17), followed by incubation with Alexa Fluor® 647 Donkey anti-rabbit IgG (Cat. No. 406414) for 1 hour at room temperature. Cells were also stained with Flash Phalloidin™ Green 488 (Cat. No. 424201) and nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured by a Revvity Operetta CLS™ High Content Analysis System with a 60X objective. Scale bar: 25 μm -
Whole cell extracts (15 μg total protein) from indicated cell lines were resolved by 3-8% Tris-Acetate gel electrophoresis, transferred to a PVDF membrane, and probed with 1:10000 dilution of Purified anti-Nestin (clone Poly22A17) for 2 hours at room temperature. Proteins were visualized by chemiluminescence detection using HRP Donkey anti-rabbit IgG (Cat. No. 406401). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Nestin is a class VI intermediate filament protein originally found in neuronal stem cells, and is required for survival and renewal of neural progenitor cells. In mixtures, Nestin forms heterodimers with vimentin or internexin, participating in the remodeling of the cell. Nestin was found to be expressed on vascular endothelial progenitor cells and various neoplasms. It is proposed to be a novel angiogenesis marker and cancer therapeutic target.

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a dilution range of 1:50 - 1:1000 is recommended. For western blotting, the suggested dilution range of this reagent is 1:5000 - 1:50000. It is recommended that the reagent be titrated for optimal performance for each application.
