    ICC staining of purified anti-PDHA1 (clone 3H2-F8-B5) on HeLa cells. Cells were co-stained with 100 nM of MitoSpy™ Red CMXRos (Cat. No. 424801) (panel A) for 20 minutes at 37°C, fixed with ice-cold methanol, then blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with purified mouse IgG1, κ isotype control (Cat. No. 400102) (panel D) or with 1.25 µg/mL anti-PDHA1 (clone 3H2-F8-B5) overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat.     IHC staining of purified anti-PDHA1 (clone 3H2-F8-B5) on formalin-fixed paraffin-embedded human cortex tissue. Following antigen retrieval using Tris-EDTA buffer (10 mM Tris, 1mM EDTA, pH 9.0), the tissue was incubated with 1 μg/mL of the primary antibody (panel B) or secondary only control (panel A) overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801) and images were captured with a 40X objective. Scale bar: 50 μm.

Description

PDHA1, pyruvate dehydrogenase E1 subunit alpha 1, is a major mitochondrial component of pyruvate dehydrogenase complexes (PDCs) which catalyzes oxidative decarboxylation of pyruvate in glycolysis and produces acetyl-CoA for the tricarboxylic acid (TCA) cycle. PDHA1 and PDHB form the heteroterameric pyruvate dehydrogenase E1 (E1) subunit of the PDC. The enzymatic activity of the PDC is regulated by phosphorylation of PDHA1 through pyruvate dehydrogenase kinase (PDK) and pyruvate dehydrogenase phosphatases (PDP). PDCs play crucial physiological functions between fed and fasting states which regulate pyruvate metabolism to maintain glucose homeostasis. PDHA1 enzymatic dysfunction causes fatal lactic acidosis and chronic neurologic dysfunction in the central nervous system. The decreased expression of PDHA1 in Parkinson’s disease and dementia with Lewy bodies patients suggests a dysfunction of mitochondrial energy production. Additionally, dysregulation of PDHA1 is found in multiple cancers, including lung, ovarian, stomach, and kidney, suggesting a role for PDHA1 in tumorigenesis.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

The suggested ICC fixation/permeabilization method is 4% paraformaldehyde plus methanol or ice-cold methanol only. Permeabilization with Triton X-100 is not recommended.

For immunohistochemistry, the suggested antigen retrieval buffer is Tris-EDTA pH 9.0.
