Total cell lysates (15 µg protein) from the indicated cell lines were resolved using 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with purified anti-PIK3R2 antibody (clone W17020A) at 1.0 µg/mL (1:500 dilution). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat-IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Equal protein loading was confirmed using Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607903) at a 1:50,000 dilution (lower). Lane M: MW ladder. Cell lysates were loaded in order of increasing predicted PIK3R2 mRNA expression levels (source: Human Protein Atlas)

Description

Phosphoinositide-3-kinase (PI3 kinase) is a lipid kinase that plays a critical role in cell signaling through generation of phosphoinositides at the plasma membrane in response to growth factor and hormone stimulation. Activation of PI3K is essential for cell growth, migration, motility, and survival. PI3K is comprised of a catalytic subunit, p110, that harbors enzymatic activity and a regulatory subunit, p85, that promotes both p110 stability and inhibition of p85. Moreover, p85 recognizes phosphorylated growth factor receptors via 2 SH2 domains that bring the PI3K dimer to the plasma membrane and in closer proximity to its substrate. Two p85 isoforms, p85α and p85β, have been described. Elevated expression of PIK3R2 (p85β) has been described in breast and colon cancers and is associated with increased tumorigenesis.

Application

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone failed to immunoprecipitate PIK3R2 from MCF-7 lysates. We therefore do not recommend W17020A for this application.

This clone failed to stain PIK3R2 from PFA-fixed and methanol permeabilized MCF-7 cells when tested by ICC. We therefore do not recommend W17020A for this application.

Due to pockets of high sequence similarity between the immunizing sequence and PIK3R1, this clone may cross-react with PIK3R1.
