    IHC staining of Purified anti-PLCγ1 (clone W23013A) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., 1X (Cat. No. 928502), the tissue was incubated without (panel A) and with (panel B) Purified anti-PLCγ1 (clone W23013A) followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective and merged. Scale bar: 50 µm

Description

Phospholipase C gamma 1 (PLCγ1) is a signaling enzyme involved in intracellular signal transduction. In response to activation of receptor-type tyrosine kinases, phospholipase C (PLC) enzymes cleave the phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2) into second messengers, namely inositol 1,4,5-triposphate (IP3) and diacylglycerol (DAG). IP3 causes the release of calcium from the endoplasmic reticulum to begin an intracellular signaling cascade, while DAG activates Protein Kinase C.

There are four families of PLCs. The PLCγ family, consisting of PLCγ1 and PLCγ2 in humans, uniquely contains SRC domains SH2 and SH3. Receptor tyrosine kinases interact with these SRC domains to enzymatically activate PLCγ1 via phosphorylation. PLCγ1 is essential for cell proliferation, differentiation, and motility. Overexpression or activation of PLCγ1 has been implicated in breast and prostate cancers as well as cancer cell invasion.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody (clone W23013A) does not cross-react with mouse PLCγ1 in Western Blot.

This antibody (clone W23013A) is not suitable for either immunocytochemistry (ICC) or intracellular flow cytometry (ICFC) due to dim signal.

For immunohistochemistry (IHC), we recommend antigen retrieval with Sodium Citrate (Cat. No. 928502). Antigen retrieval with Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0) is not recommended due to non-specific staining.
