Whole cell extracts (15 µg total protein per lane) from indicated human cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-TATA Binding Protein (clone W22002D) overnight at 4°C (left blot). For comparison, we probed a separate membrane (right blot) containing the same human lysates with another antibody on the market, Purified anti-TATA Binding Protein (clone 1TBP18). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of Purified rat IgG2a, κ isotype control (Cat. No. 400502) or Purified anti-TATA Binding Protein (clone W22002D). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with a control antibody (clone 1TBP18) (Cat. No. 668306) against a separate epitope of anti-TATA Binding Protein. Lane M: Molecular weight marker -
HCT 116 cells were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-TATA Binding Protein (clone W22002D) (panel A), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801) (panel B). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged (panel C). Scale bar: 50 µm -

The TATA-binding protein, also known as TBP or Tata Binding Protein, is the general transcription factor that functions at the core of the DNA-binding multiprotein factor TFIID. Binding of TFIID to the TATA box is the initial transcriptional step of the pre-initiation complex (PIC), which plays a role in the activation of eukaryotic genes transcribed by RNA polymerase II. This protein is a component of the transcription factor SL1/TIF-IB complex, which is involved in the assembly of the PIC during RNA polymerase I-dependent transcription. The rate of PIC formation is assumed to be primarily dependent on the rate of the association of SL1 with the rDNA promoter. SL1 is involved in stabilization of nucleolar transcription factor 1/UBTF on rDNA.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For the use of this antibody (clone W22002D) in immunocytochemistry (ICC), we recommend fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with 100% ice-cold methanol.
