In response to viral infection, plasmacytoid dendritic cells (pDCs) produce interferons, most notably type I and type II. As the anti-viral immune response is a tightly regulated process, Liu et al. studied how a microRNA can modulate cytokine production in response to TLR stimulation. Expression of microRNA-21 is induced in pDCs following TLR stimulation and deletion of this microRNA results in impaired production of both IFN-α and IFN-γ. Investigation into the mechanism by which microRNA-21 induces cytokine release revealed that it suppresses the P13K/MTOR signaling pathway through targeting PTEN, resulting in increased expression and nuclear translocation of the IRF7 transcription factor driving interferon expression. These studies implicate a role and potential target for microRNAs in modulating immune responses.

MojoSort™ Mouse Pan Dendritic Cell Isolation Kit


A single cell suspension from C57BL/6 mouse splenocytes was prepared using enzymatic digestion to isolate Pan Dendritic Cells (classical and plasmacytoid) using the MojoSort™ Mouse Pan Dendritic Cell Isolation Kit. Cells were stained with anti-mouse CD11c (clone N418) PE and anti-mouse CD317 (clone 927) APC. Dead cells were excluded by Helix NP™ Blue.
