Recombinant mouse IL-13 (Cat. No. 575902) (black circles) induces proliferation on human TF-1 cells. LEAF™ purified anti-mouse IL-13 (clone Poly5229) (purple squares) neutralizes the cell proliferation induced by recombinant mouse IL-13 at 15 ng/mL in human TF-1 cells in a dose-dependent manner, whereas the LEAF™ purified goat IgG isotype control (gray triangles) does not have the effect. ND50 range: 0.08 - 0.6 μg/mL. -
Whole cell extracts (15 µg total protein) from HeLa cells spiked with the indicated amount of recombinant mouse IL-13 protein (Cat. No. 575908) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.1 µg/mL LEAF™ purified anti-mouse IL-13 (clone Poly5229) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-goat. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Mouse IL-13 was initially cloned from cDNA libraries of activated T cells and was designated as P600. IL-13 is an immunoregulatory cytokine secreted predominantly by activated T(H)2 cells, and it is a key mediator in the pathogenesis of allergic inflammation. IL-13 shares many functional properties with IL-4, and they share a common receptor subunit, the alpha subunit of the IL-4 receptor (IL-4Rα). IL-13 mediates its effects by interacting with a complex receptor system comprised of IL-4Rα and two IL-13 binding proteins, IL-13Rα1 and IL-13Rα2. Ligation of the IL-13 receptor complex results in signaling via the insulin receptor substrate (IRS)-1 and 2 and STAT-6 pathways. Interleukin-13 (IL-13), like IL-4, is a cytokine produced by T(H)2 type helper T cells in response to signaling through the T cell antigen receptor and by mast cells and basophils upon cross-linkage of the high-affinity receptor for immunoglobulin E (IgE). IL-13 has been implicated in airway hypersensitivity and mucus hypersecretion, inflammatory bowel disease, and parasitic nematode expulsion.

Each lot of this antibody is quality control tested by neutralizing proliferation induced by recombinant mouse IL-13 at 15 ng/mL in human TF-1 cells. ND50: 0.08 - 0.6 μg/mL. For western blotting, the suggested use of this reagent is 0.02 - 0.5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
