Lysosomes are cellular organelles involved in degrading cellular debris during autophagy, as well endocytosed material. Defects in lysosomal function are related to neurodegenerative disorders, cancers, and aging-related diseases. Lysosome Probe Red labels lysosomes in live cells and selectively accumulates in lysosomes via the lysosomal pH gradient. Fluorescence is greatly enhanced upon entry into lysosomes. Its photostability and excellent cellular retention makes this probe particularly useful for live cell imaging applications, as well as for flow cytometry.

Component:
20 μL of 500X Lysosome Probe Red (-20°C, protected from light, avoid freeze/thaw cycles)
Required Materials Not Included:
Buffer: PBS, phenol-free media, or HBSS
Detection/Imaging Guidelines:
Ex/Em = 575/597 nm
Fluorescence microscope filter set: Texas Red
Flow cytometry channel: PE/Cyanine 7 with yellow/green laser excitation
Assay Protocol:- Thaw the 500X Lysosome Probe and allow it to reach room temperature.
- Prepare lysosome staining solution by diluting the Lysosome Probe 1:500 in your buffer of choice.
Note: Probe concentration may need to be adjusted according to cell type and assay conditions. - Optional: Include live-cell nuclear counterstain such as DRAQ5™ (Cat. No. 424101).
- Add an equal volume of Lysosome Staining solution to cell culture media.
- Incubate the cells at 37°C for 30 minutes.
- Wash cells by twice with PBS or HBSS. If cells are in suspension, spin down and resuspend in buffer of choice.
- Add desired volume of PBS or HBSS to cells for observation.
- Proceed immediately to analysis.
