A single-cell suspension mixture of 30% human peripheral blood mononuclear cells (PBMCs) with 70% human colorectal adenocarcinoma cell line HT-29 was prepared for positive selection of CD45+ cells using the MojoSort™ Human CD45 Selection Kit. Cells were stained with APC anti-human CD326 (clone CO17-1A) (Cat. No. 369809) and a cocktail of PE conjugated antibodies against CD3/CD14/CD15/CD19/CD56 (clones UCHT1/HCD14/W6D3/HIB19/HCD56) (Cat. Nos. 300407/325605/323005/302207/318305). Dead cells were excluded by Helix NP™ Blue (Cat. No. 425305).

Human CD45+ expressing cells are either selected or depleted by incubating the sample with anti-human CD45 Biotin Antibody followed by incubation with magnetic Streptavidin Nanobeads. The magnetically labeled fraction is retained by the use of a magnetic separator. After collection of the CD45+ cells, downstream applications include functional assays, gene expression, phenotypic characterization, etc.
MojoSort™ reagents are also compatible with column-based cell separation systems available from other vendors. Optimized protocols for cell separation using columns from in-house testing are provided for each kit under the “Related Protocols” section, as well as representative data on the product webpage (where available). Data generated using column separators are indicated on the figure legend.
Due to the property of the beads, MojoSort™ reagents typically require dilution for optimal use on column separators. Where available, recommended dilution factors for each kit component based on in-house testing are provided under the “Application Notes” section of the webpage
