    IHC staining of purified anti-human CD90 (Thy1) (clone S22015F) on formalin-fixed paraffin-embedded human kidney (A) or tonsil (B, negative control) tissue. After antigen retrieval using 1X Citrate Buffer (Cat. No. 420902), the tissues were incubated with 10 μg/mL of purified anti-human CD90 (Thy1) (clone S22015F) at 4°C overnight, followed by incubation with 2.5 μg/mL of Alexa Fluor® 594 Goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 10X objective.

Description

CD90, also known as Thy-1, is a 25-35 kD GPI-anchored protein. It belongs to the Ig superfamily. Human CD90 is expressed on neuronal cells, a subset of CD34+ cells, a subset of fetal liver cells and fetal thymocytes, fibroblasts, activated endothelial cells, and some leukemia cell lines. CD34+CD90+ cells are primitive hematopoietic stem cells. It has been reported that Thy-1 binds with β2 and β3 integrins and plays bimodal roles in the regulation of cell adhesion and neurite outgrowth, and inhibits hematopoietic stem cells proliferation and differentiation.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration of 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone S22015F does not block the binding of clones S22015B nor 5E10 on target cells.
