    Total lysates (15 µg protein) from HeLa (Human), Raw264.7 (Mouse) and UMR106 (Rat) were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:5000 purified Vimentin antibody, clone Poly29191. Proteins were visualized using chemiluminescence detection by incubation with HRP Donkey anti-chicken secondary antibody (1:3000 dilution). Direct-Blot™ HRP anti-β-actin was used as a loading control (Cat. No. 643807, 1:8000 dilution).

    Total cell lysates (15 µg total protein) from Daudi (negative control), PC-3, Jurkat and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 0.1 µg/mL (1:5,000 dilution) of Purified anti-Vimentin Antibody, clone Poly29191, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP conjugated donkey anti-Chicken IgY antibody. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

    Immunofluorescence of A431 cells with (A) Chicken IgY isotype control (Negative, Cat. No. 402101) or (B-D) Vimentin primary antibody (clone Poly29191). Alexa Fluor® 488 (Green) Goat anti-Chicken IgY was used as secondary antibody. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective using KEYENCE BZ-X700 fluorescence microscope. Exposure time (Seconds) for (A) is 1/15, and (B-D) is 1/70. Concentrations for (A, B) is 1/1000, (C) is 1/2500, (D) is 1/5000.

Description

Vimentin is a protein subunit of the 10nm or intermediate filaments protein found in many kinds of mesenchymal and epithelia cells, several types of cells in tissue culture and in developing neuronal and astrocytic precursor cells in the central nervous system. Vimentin frequently forms copolymers with other intermediate filament proteins, such as GFAP (in many kinds of astrocytes), with desmin (in muscle cells) and neurofilament proteins (in developing neurons). Antibodies to vimentin are useful in studies of stem cells and generally to reveal the filamentous cytoskeleton.

Recommended Usage

Each lot of this antibody is quality control tested by immunohistochemistry. For immunohistochemistry, a dilution range of 1:200 - 1:500 is suggested. For Western blotting, the suggested use of this reagent is 1:2000 - 1:8000 dilution. For immunocytochemistry, a dilution range of 1:1000 - 1:5000 is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in immunofluorescence (IF) and immunohistochemistry (IHC). This antibody does not bind to protein A or protein G.
