Description

Interleukin-1 receptor antagonist (IL-1RA or IL-RN) is a member of IL-1 family cytokine and it is a naturally occurring inflammatory inhibitor protein.  It competes with IL-1α and IL-1β for binding to type I and type II receptors.  It can modulate a variety of interleukin 1 related immune and inflammatory responses. IL-1RA has been shown to block the inflammatory responses induced by IL-1 both in vitro and in vivo model. Recombinant IL-1RA (anakinra) has been used as to treat rheumatoid arthritis. A polymorphism of IL-1RN gene is reported to be associated with increased risk of osteoporotic fractures, gastric cancer, bladder cancer, and other cancer types.

Human IL-1R antagonist (IL-1RA), amino acids (Arg26-Glu177) (P18510), was expressed in E. coli.
Molecular Mass
The 153 amino acid N-terminal methionylated recombinant protein has a predicted molecular mass of 17.2 kD. The predicted N-terminal amino acid is Met.
Purity
>98%, as determined by Coomassie stained SDS-PAGE and HPLC analysis.
Formulation
Lyophilized, carrier-free.
Endotoxin Level
Less than 0.1 ng per µg of protein.
Storage & Handling
Unopened vial can be stored at -20°C or -70°C. For maximum results, quick spin vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. It is recommended to further dilute in a buffer, such as 5% Trehalose, and store working aliquots at -20°C to -80°C. Avoid repeated freeze/thaw cycles.
Activity
ED50 = 7 - 35 ng/ml, corresponding to a specific activity of 0.2 - 1.4 x 105 units/mg, as determined by inhibition of D10.G4.1 cell proliferation induced by human IL-1α.
