Recombinant human PDGFRα Chimera was aliquoted in PBS at 0.2 mg/mL and one aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). After this procedure, the samples were tested for their ability to inhibit the proliferation of NIH3T3 in the presence of the human PDGF-AA in a dose-dependent manner. The ED50 for this effect is 0.4 – 2.4 µg/mL

Description

The Platelet-derived growth factor (PDGF) family comprises four different polypeptide chains encoded by four genes. The four PDGF chains assemble into disulfide-bonded dimers via homo- or heterodimerization, and five different dimeric isoforms have been described so far; PDGF-AA, PDGF-AB, PDGF-BB, PDGF-CC, and PDGF-DD. PDGF is a major mitogen for fibroblasts, smooth muscle cells, and others. They exert their functions by interacting with two related receptor tyrosine kinases, PDGF receptor α (PDGFRα) and β, and the PDGFRα as a universal receptor. All the currently described PDGF receptor-binding components, e.g. phospholipase C-gamma, members of the src family of cytoplasmic tyrosine kinases, the rasGTPase activating protein and p85, the regulatory subunit of phosphatidylinositol 3' kinase, contain a conserved src homology 2-domain, through which the association with the receptor takes place. The receptor-binding components either possess an intrinsic enzymatic activity or function as adaptors, which may be complex with catalytically active components. PDGFRα plays a key role in many diseases through the above signal transduction.

Human PDGFRα, amino acid (Gln24-Glu524) (Accession: #P16234.1), with a linker (IEGRMD) and a C-terminal human IgG (Pro100-Lys330), was expressed in CHO cells.
Molecular Mass
The 738 amino acid recombinant protein has a predicted molecular mass of approximately 82.8 kD. The DTT-reduced and non-reduced protein shall migrate at approximately 85 -120 kD and 140 – 240 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Gln.
Purity
> 95%, as determined by Coomassie-stained SDS-PAGE.

0.22 µm filtered protein solution is in PBS, pH 7.2
Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
