Rat_IL-9_RECOM_CF_1_050924
Rat IL-9 induces proliferation of MC/9 mouse mast cells in a dose dose-dependent manner. The ED50 for this effect is 80 - 800 ng/mL.
  • Rat_IL-9_RECOM_CF_1_050924

  • Rat_IL-9_RECOM_CF_2_050924
Stability Testing for Recombinant Rat IL-9. Recombinant rat IL-9 was aliquoted in PBS and pH 7.2. at 0.2 mg/mL. One aliquot was kept at 4°C (control), and another was frozen and thawed four times (4x Freeze/Thaw). The samples were tested by their ability to induce the proliferation of MC/9 mouse mast cells in a dose-dependent manner. The ED50 for this effect is 80 – 800 ng/mL.

Description

Interleukin-9 (IL-9) regulates variety of hematopoietic cells, stimulates cell proliferation, and prevents apoptosis. Mature rat IL-9 shows 58% and 74% amino acid sequence homology with human and mouse IL-9, respectively. The IL-9 receptor consists of the cytokine-specific IL-9 receptor α-chain (IL-9Rα) and the common γ-chain that is shared by IL-2, IL-4, IL-7, IL-15 and IL-21. IL-9 is expressed by activated Th9, Th17, Treg, and mast cells and appears to play important roles in the development and function of subsets within the CD4+ T cell lineage. IL-9 also inhibits immune responses by enhancing the suppressive properties of Tregs and by recruiting immune suppressive mast cells to sites of inflammation. Genetic studies on a mouse model of asthma demonstrated that IL-9 plays a key role in the pathogenesis of bronchial hyperresponsiveness correlating with the observed increased expression of IL-9 and IL-9R in lungs of asthmatic patients. In addition, IL-9 promotes mast cell mediated intestinal permeability, development of food allergies and atopic dermatitis.

Product Details

Source
Recombinant rat IL-9, amino acid Gln19-Ala144 (Accession No. D4A819) was expressed in 293E.
Molecular Mass
The 126 amino acid recombinant protein has a predicted molecular mass of approximately 14.1 kD. The DTT-reduced and non-reduced protein migrate at approximately 27 - 35 kD by SDS-PAGE. The predicted N-terminal amino acid is Gln.
Purity
> 95%, as determined by Coomassie stained SDS-PAGE.
Formulation
0.22 µm filtered protein solution is in PBS, pH7.2
Endotoxin Level
Less than 0.1 EU per µg protein as determined by the LAL method.
Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
ED50 = 80 – 800 ng/mL, as determined by a dose-dependent stimulation in a proliferation assay with MC/9 cells. Deep Blue Cell Viability™ Kit (Cat. No. 424701) is used to measure the proliferation.
Application

Bioassay

Application Notes

BioLegend carrier-free recombinant proteins provided in liquid format are shipped on blue-ice. Our comparison testing data indicates that when handled and stored as recommended, the liquid format has equal or better stability and shelf-life compared to commercially available lyophilized proteins after reconstitution. Our liquid proteins are validated in-house to maintain activity after shipping on blue ice and are backed by our 100% satisfaction guarantee.
