Murine B-cells were isolated using the MojoSort™ Mouse Pan B Cell Isolation Kit (Cat. No. 480051) and activated overnight with 20 ng/mL recombinant mouse IL-4 (carrier-free) (Cat. No. 574302) and 10 µg/mL purified anti-mouse IgM (Cat. No. 406501). Activated B cells were seeded onto tissue culture plates pre-coated with 20 µg/mL Ultra-LEAF™ purified anti-mouse CD44 (clone W19210A) or isotype control. After 4 hours, the number of elongated cells (circularity < 0.5) was quantified using brightfield imaging and morphometric analysis with ImageJ. The assay was performed in triplicate, and 4 images per well at 20x magnification were analyzed. Data shown are normalized to rat IgG2a, κ isotype control treated wells.

After 4 hours, cells were fixed with Fixation Buffer (Cat. No. 420801) and stained with Flash Phalloidin™ Green 488 (Cat. No. 424201) in Intracellular Staining Permeabilization Wash Buffer (10X) (Cat. No. 421002). Images were taken using a 40x objective.

Description

CD44 is an 80-95 kD glycoprotein also known as Hermes, Pgp1, H-CAM, or HUTCH. It is expressed on all leukocytes, endothelial cells, hepatocytes, and mesenchymal cells. As B and T cells become activated or progress to the memory stage, CD44 expression increases from low or mid levels to high levels. Thus, CD44 has been reported to be a valuable marker for memory cell subsets. High CD44 expression on Treg cells has been associated with potent suppressive function via high production of IL-10. CD44 is an adhesion molecule involved in leukocyte attachment to and rolling on endothelial cells, homing to peripheral lymphoid organs and to the sites of inflammation, and leukocyte aggregation.

Concentration
The antibody is bottled at the concentration indicated on the vial, typically between 2 mg/mL and 3 mg/mL. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C. This Ultra-LEAF™ solution contains no preservative; handle under aseptic conditions.

Activ - Quality tested

Recommended Usage

Each lot of this clone is quality control tested by induction of spreading of activated murine B-cells on a tissue culture plate coated with 20 µg/mL anti-mouse CD44. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone activates CD44 on murine B-cells as shown by induction of B-cell spreading on anti-mouse CD44 coated surface.
