A20103J_ULEAF_SARS-CoV-2-S_Protein-S1_Antibody_021521
Recombinant human ACE2 (Cat. No. 792002) (black circles) binds to immobilized recombinant SARS-CoV-2 S protein S1-Fc chimera (Cat. No. 793004) in a dose-dependent manner. Purified anti-SARS-CoV-2 S protein S1 antibody (clone A20103J) (purple squares) inhibits the binding in a dose-dependent manner. This antibody blocks the binding of 0.5 µg/mL recombinant human ACE2 to 1.0 µg/mL immobilized recombinant SARS-CoV-2 S protein S1-Fc chimera. ND50 range: 0.04 - 0.2 µg/mL.

Description

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a single-stranded RNA virus that belongs in a family of viruses known as coronaviruses. SARS-CoV-2 infection, known as COVID-19, was declared a pandemic by WHO on March 11, 2020 and causes symptoms leads to respiratory infection, pulmonary failure which can be fatal. SARS-CoV-2 is structurally composed of 4 main proteins (e.g. spike glycoprotein, envelope glycoprotein, membrane glycoprotein and nucleocapsid protein) and several accessory proteins. The spike glycoprotein (S) is a transmembrane molecule that forms homotrimers on the surface of the virus and facilitates virulence through binding to angiotensin-converting enzyme 2 (ACE2) expressed on host cells. This protein is composed of two subunits S1 and S2. S1 is responsible for binding to the host cell via ACE2 receptor and the S2 subunit is responsible for fusion with the host cell.

Blocking - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding of 0.5 µg/mL recombinant human ACE2 (Cat. No. 792002) to 1 µg/mL immobilized recombinant SARS-CoV-2 S protein S1-Fc chimera (Cat. No. 793004). ND50 range: 0.04 - 0.2 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
