Different amounts of V5 tagged proteins (Genescript, M0101) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified monoclonal V5-tag (clone 7/4) antibody. Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP and chemiluminescence detection.

200ng V5 tagged protein was resuspended in PBS. Lane 1 was immunoprecipitated with 2 µg mouse IgG. Lane 2 was immunoprecipitated with 2 µg anti-V5-tag (clone 7/4) antibody. Lane 3 was immunoprecipitated with 2 µg anti-V5-tag (clone poly290238) antibody. Lane 4 was loaded with 50ng V5 tagged protein directly as a positive control. Western blot analysis was performed using mouse anti-V5 tag antibody (clone 7/4) and followed by HRP-anti-mouse IgG.

Description

Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. The V5 tag is a 14 amino acid peptide derived from a small epitope (Pk) on the P and V proteins of simian virus 5 (SV5), a member of the paramyxovirus family. Because of its small size, the addition of a V5 tag sequence (GKPIPNPLLGLDST) to a given gene creates a stable fusion product that does not appear to interfere with the biochemical properties of the protein. Many recombinant proteins have been engineered to express a short sequence derived from the V5 molecule known as V5-tag.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 0.5 µg per ml. For immunoprecipitation, the suggested use of this reagent is 1-5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
