MojoSort on Columns™ Cell Separation System is BioLegend’s new portfolio of reagents optimized for column-based magnetic cell separation. Our cell separation columns contain an iron sphere matrix, which generate a strong magnetic field when placed on BioLegend’s Column, Separator, and Multistand. Our column system can be used for positive or negative selection of MojoSort on Columns™ kits and reagents. In negative selection, the untouched cells flow through the column and are collected directly into a tube. In positive selection, the labeled cells are retained in the column matrix and eluted by removing the column from the magnetic separator.

 

MojoSort on Columns™ reagents and kits are optimized for use on our columns, separators, and stands. They have also demonstrated compatibility with third party columns, column separators and stands. Please contact BioLegend Technical Service for more guidance on compatibility with third party systems.

MojoSort on Columns™ Kit Comparisons

 

Data comparing BioLegend MojoSort on Column kits with competitors' reagents for CD4 and NK cell isolation.

 

(Top) Human CD4 T cells were isolated from fresh PBMCs using the MojoSort on Columns™ Human CD4 T Cell Isolation Kit with a BioLegend Large Column and Separator. For comparison, we simultaneously performed cell separation with a competitor’s equivalent column system and column kit for human CD4 T cells. The negatively selected cells were then stained with Spark Violet™ 538 anti-CD3 and APC anti-CD4. Then the cells were analyzed by flow cytometry on CD45+ live cells.

 

(Bottom) Untouched NK cells were isolated from frozen Leukopak-derived PBMCs using the MojoSort on Columns™ Human NK Cell Isolation Kit with BioLegend’s Large Columns and Separator. The negatively selected cells were then stained with APC anti-human CD3 and PE anti-human CD56. Then, the cells were analyzed by flow cytometry gated on live CD45+ cells.​

Untouched CD4+ T cells were isolated from mouse splenocytes using BioLegend’s Large Columns and equivalent medium flow columns from a competitor. The negatively selected cells were than stained with FITC anti-mouse CD45, PE anti-mouse CD4, APC anti-mouse CD3, and Helix NP™ Blue. Then, the cells were analyzed by flow cytometry gated on live CD45+ cells to determine purity and yield.
