    C57BL/6 mouse frozen brain tissue was fixed with 4% paraformaldehyde (PFA) for ten minutes, permeabilized with 0.5 % Triton X-100 for ten minutes, and blocked with 5% FBS for 1 hour. Then the tissue was stained with 5 µg/mL of Alexa Fluor® 488 anti-Tubulin Beta 3 (TUBB3) (Clone AA10) (shown in green) and 5 µg/mL of Alexa Fluor® 594 anti-GFAP (Clone 2E1.E9) (shown in red) in blocking buffer, overnight at 4°C. Nuclei were counterstained with DAPI (blue). The image was captured with 10X objective.

    Dissected C57/B6 mouse small intestine was immersed in 4% paraformaldehyde (PFA) overnight followed by 30% sucrose immersion overnight and frozen in OCT. Frozen section was blocked with 5% FBS and 5% mouse serum for 30 minutes at room temperature. Then the tissue section was stained with 2.5 µg/mL of anti-mouse Tubulin Beta 3 (clone AA10) Alexa Fluor® 488 (green) and 2.5 µg/mL of anti-mouse CD326 (clone G8.8) overnight at 4°C. Nuclei were counterstained with DAPI (blue). The image was captured by 10X objective.

    Formalin-fixed, 300 micron-thick mouse kidney section was blocked, permeabilized and stained overnight with CD31 (clone MEC13.3) Alexa Fluor® 647 (red) at 5 µg/mL, Tubulin Beta 3 (TUBB3) (clone AA10) Alexa Fluor® 488 (green) at 2.5 µg/mL, and Podoplanin (clone Pmab-1) Alexa Fluor® 594 (blue) at 5 µg/mL, optically cleared, then analyzed at 230 µm imaging depth on a confocal microscope. Description

Tubulin is the main component of microtubules. In adults, tubulin beta 3 (TUBB3) is primarily expressed in neurons and is commonly used as a neuronal marker. It plays an important role in neuronal cell proliferation and differentiation. Mutations in this gene cause congenital fibrosis of the type 3 extraocular muscles. Tubulin beta 3 (TUBB3) is also found in a wide range of tumors. Studies indicate that it is a predictive and prognostic marker in various tumors.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. For immunohistochemical staining on frozen tissue sections, the suggested use is 1.25 - 5.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 488 has a maximum emission of 519 nm when it is excited at 488 nm.
