HeLa cells were fixed with 1% paraformaldehyde (PFA) and blocked with 5% FBS for 30 minutes. Then the cells were stained with 10 µg/ml anti-human EGFR (clone AY13) Alexa Fluor® 594 (red) at room temperature for 3 hours in dark. Nuclei were counterstained with DAPI (blue). The image was captured with a 40X objective.

    Formalin-fixed paraffin-embedded human breast cancer tissue slices were deparaffinized and rehydrated. Antigen retrieval was done with Tris-Buffered Saline 1X (1.0M, pH7.4) at 95°C for 40 minutes, washed with PBS/ 0.05% Tween20 twice for five minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the slices were stained with 10 µg/ml of Alexa Fluor® 594 anti-human EGFR (clone AY13) (red) at 4°C overnight. Nuclei were counterstained with DAPI (blue). The image was captured with a 10X objective.

Description

Epidermal growth factor receptor (EGFR) is a transmembrane glycoprotein and member of the protein kinase superfamily that regulates cell growth and differentiation. EGFR binds EGF, TGF-α, amphiregulin, betacellulin, heparin-binding EGF-like growth factor, GP30, and vaccinia virus growth factor - all members of the EGF family. Ligand binding induces EGFR dimerization and autophosphorylation, initiating the MAPK, Akt, and JNK signaling pathways. EGFR is expressed by epithelial and endothelial cells and is frequently expressed by epithelial carcinomas.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 2.5 - 10 μg/mL is recommended. For immunohistochemical staining on formalin-fixed paraffin-embedded tissue sections, the suggested use of this reagent is 5.0 - 10 µg per mL. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 594 has an excitation maximum of 590 nm, and a maximum emission of 617 nm.
