Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) purified anti-acetyl-CoA carboxylase 1 antibody (clone W19216A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker. -
HeLa cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with purified rat IgG2a, κ isotype ctrl antibody (Cat. No. 400502) (panel A) or 1.0 µg/mL purified anti-acetyl-CoA carboxylase 1 antibody (clone W19216A) (panel B) followed by incubation with Alexa Fluor® 594 goat anti-rat IgG antibody (Cat. No. 405422) at 2.0 µg/mL (1:250 dilution). Nuclei were counterstained with DAPI, and the image was captured with a 60X objective. -
Whole cell extracts (250 μg total protein) prepared from Hep G2 cells were immunoprecipitated overnight with 2.5 μg purified rat IgG2a, κ isotype ctrl antibody (Cat. No. 400502) or purified anti-acetyl-CoA carboxylase 1 antibody (clone W19216A). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with W19216A. Lane M: Molecular weight marker.

Acetyl-CoA carboxylase (ACC) is a cytosolic enzyme that catalyzes carboxylation of acetyl-CoA to malonyl-CoA, the initiating and rate-limiting step in fatty acid biosynthesis. Two isoforms of ACC exist that show tissue-specific distribution. ACC1 is predominantly expressed in lipogenic tissues, while ACC2 is found in oxidative tissues. In response to depletion of cellular ATP, AMPK phosphorylates ACC1 at Ser79, resulting in loss of enzymatic activity. ACC1 is a clinical target for various metabolic diseases, and also contributes to metabolic reprogramming in tumor cells.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 2.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

-This clone was tested for ICC using 4% PFA-fixed HeLa cells permeabilized with either Triton X-100 or methanol. Both methods were compatible with staining of acetyl-CoA carboxylase 1.
