Total cell lysates (15 µg total protein) from HEK293 and Daudi (negative controls), HaCat and A431 (positive controls cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL of Purified anti-CA125 Antibody, clone 618F, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. -
IHC staining of anti-CA125 (clone 618F) antibody on formalin-fixed paraffin-embedded human ovarian tumor. Following antigen retrieval using Retrieve-All Antigen Unmasking System's Retrieve-All 1 (Universal pH 8.0), the tissue was incubated with the primary antibody at 0.5 µg/ml for 60 minutes at room temperature. BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit was used for detection, followed by counterstaining with hematoxylin.

CA125 is believed to provide a protective, lubricating barrier against particles and infectious agents at mucosal surfaces. It binds to MSLN, which mediates heterotypic cell adhesion. This may contribute to the metastasis of ovarian cancer to the peritoneum by initiating cell attachment to the mesothelial epithelium through its binding to MSLN.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.5 µg per ml. For immunohistochemistry, a concentration range of 0.5 - 5 µg per ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

The observed molecular weight of this target is ~500 kD, which is considerably different than the predicted MW of 1519 kD. Similar observed molecular weights were confirmed using control antibodies from other vendors and have been reported by other vendors.
