Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-DNM1L (clone W20081E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:25000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker -
ICC staining of purified anti-DNM1L (clone W20081E) on HeLa cells. The cells were fixed and permeabilized with ice cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 2.5 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-rat IgG (Cat. No. 405422) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801), and the image was captured with a 60X objective.

Dynamin-1-like protein (DNM1L) is a member of the dynamin superfamily of GTPases. Its best studied role is in the control of mitochondrial fission. DNM1L oligomerizes into filaments that form rings around the scission site to “pinch off” dividing mitochondria using GTP hydrolysis. Accordingly, DNM1L also has important functions in the mitochondrial dynamics of cell proliferation and apoptosis. DNM1L is also required for proper formation of the nervous system, and deficiencies in its functions have been linked to Alzheimer’s Disease.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 2.5 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For Western Blot, higher molecular weight band (≈100 kDa) likely corresponds to SUMOylated DNM1L (e.g. Figueroa-Romer, et al. 2009).
For ICC, we recommend fixation/permeabilization with 100% ice cold methanol alone. Alternatively, 4% paraformaldehyde fixation followed by permeabilization with ice cold methanol is possible. We do not recommend 4% paraformaldehyde fixation followed by permeabilization with 0.5% Triton-X, due to incorrect signal localization.
