A17096A_PURE_DOPA_Antibody_1_090919
Western blot of purified anti-DOPA Decarboxylase (DDC) antibody (clone A17096A). Lane 1: Molecular weight marker; Lane 2: 30 µg of mouse brain lysate; Lane 3: 20 µg of HEK293 cell lysate. The blot was incubated with 5 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

IHC staining of anti-DOPA Decarboxylase (DDC) antibody (clone A17096A) on formalin-fixed paraffin-embedded mouse brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R, the tissue was incubated with 0.1 µg/ml of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Description

DOPA decarboxylase (DDC) is the enzyme that catalyzes several decarboxylation reactions including the conversion of L-3,4-dihydroxyphenylalanine (DOPA) to dopamine, L-5-Hydroxytryptophan (5-HTP) to serotonin, and L-tryotophan to tryptamine. DDC is the rate-limiting enzyme in dopamine synthesis in patients treated with L-DOPA and in serotonin synthesis in people treated with 5-HTP, such as those affected by Parkinson’ disease and Depression, respectively. Therefore, DDC is an important therapeutic target for these disorders. DDC deficiency is associated with severe developmental delay, oculogyric crises, and autonomic dysfunction.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 5.0 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10 µg/ml in human and 0.05 - 10 µg/ml for mouse is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
