Western blot of purified anti-Gemin 4 antibody (clone 17D10). Lane 1: Molecular weight marker; Lane 2: 10 µg of HEK293 cell lysate; Lane 3: 10 µg of HepG2 cell lysate. The blot was incubated with 2 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system. Note that expression of Gemin 4 in HEK293 cells is much higher than in HepG2 cells.

Description

Gemin 4 is one of the core components of the survival of motor neurons (SMNs) macromolecule protein complex, which plays an important role in the assembly and re-structuring of small nucleoproteins and transcriptosomes. Recent studies have shown that one of the three putative nuclear localization signal motifs in Gemin 4 is necessary and sufficient to direct the nuclear import of the other two SMN complex components, Gemin 2 and Gemin 3. This suggests that Gemin 4 plays an essential role in mammalian snRNP biogenesis via facilitating the import of the SMN complex or subunits into the nucleus.
