Whole cell extracts (15 μg total protein per lane) from indicated cell lines were resolved by 4-20% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-Histone H3 (C-terminus) Recombinant (clone 1B1-B2.recM1) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining of Purified anti-Histone H3 (C-terminus) (clone 1B1-B2.recM1) on formalin-fixed paraffin-embedded human colon tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated without (panel A) and with (panel B) Purified anti-Histone H3 (C-terminus) Recombinant (clone 1B1-B2.recM1) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. The tissue was subsequently co-stained with Alexa Fluor® 488 anti-Vimentin (clone O91D3) (Cat. No.677809) (green) and nuclei were counterstained with DAPI (Cat. No. 422801) (blue). Images were captured with a 40X objective and merged. Scale bar: 50 μm -
HeLa cells were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained either with Purified Mouse IgG1, κ Isotype Control (panels A and C) or Purified anti-Histone H3 (C-terminus) Recombinant (clone 1B1-B2.recM1) (panels B and D), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) (red) for 1 hour at room temperature. Cells were subsequently co-stained with Alexa Fluor® 488 anti-Tubulin-α (clone 10D8) (Cat. No. 627906) (green) and nuclei were counterstained with DAPI (Cat. No. 422801) (blue). Histone proteins are classified into core histones (H2A, H2B, H3, H4) and linker histones (H1, H5). Core histones form an octamer, which contains two H2A-H2B dimers and one H3-H4 tetramer. Core histones are predominantly globular except for the unstructured N-terminal tails. Posttranslational modifications, such as acetylation, methylation, phosphorylation, ubiquitination, SUMOylation and ADP-ribosylation occur in histone tails.
Histone modifications induce changes of chromatin structure and thereby affect the accessibility of transcription factors, nuclear proteins and enzymes to genomic DNA, resulting in gene activation or repression. It is known that histone modifications play critical roles in DNA repair, DNA replication, transcription regulation, alternative splicing and chromosome condensation and some diseases including autoimmune diseases and cancers.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.0125 - 0.025 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 0.25 - 1.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 0.5 - 1.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For immunohistochemistry use on formalin-fixed paraffin-embedded tissue (IHC-P), either antigen retrieval with Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704) is recommended.
For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:- Fixation buffer (Cat. No. 420801) followed by 0.5 % Triton X-100
- Fixation buffer (Cat. No. 420801) followed by 100% ice-cold methanol
- 100% ice-cold methanol only
