Human peripheral blood mononuclear cells were treated with True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) then stained with purified anti-human Bcl11b (clone W17189A) (left) or purified rat IgG2a, κ isotype control (right) antibody, followed by anti-rat IgG PE. Cells were stained for anti-human CD3 APC. Data was gated on lymphocyte populations. -
Jurkat cells were treated with True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401) then stained with purified anti-human Bcl11b (clone W17189A) (filled histogram) or purified rat IgG2a, κ isotype control (open histogram), followed by anti-rat IgG PE. -
Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.25 µg/mL (1:2000 dilution) of purified anti-human Bcl11b (clone W17189A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin (Cat. No. 664804) was used as a loading control at a 1:10000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining using purified anti-human Bcl11b (clone W17189A) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using 1X Tris-Buffered Saline (final concentration 0.05M) with Tween-20 (Cat. No. 925501), the tissue was incubated with 5 µg/mL of the antibody (left) or left untreated (right) overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-rat IgG (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scalebar = 50 µM

The transcription factor, Bcl11b, is expressed in the thymus, mainly T cells. It is indispensable for T lineage development. When Bcl11b was deleted, T cells from all developmental stages acquired NK cell properties and concomitantly lost or decreased T cell-associated gene expression. These induced T-to-natural killer (ITNK) cells, which were morphologically and genetically similar to conventional NK cells, killed tumor cells in vitro, and effectively prevented tumor metastasis in vivo. Therefore, ITNKs may represent a new cell source for cell-based therapies.

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
