Total cell lysates (15 µg total protein) from A549 and HEL (negative controls) and HaCaT (positive control) cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 µg/mL of Purified anti-KRT1 Antibody, clone Poly19052, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-rabbit IgG Antibody (Cat. No. 406401) at a 1:3000 dilution. Direct-Blot™ HRP anti-β actin Antibody (Cat. No. 664804) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker. Predicted KRT1 expression data obtained from Human Protein Atlas.

    IHC staining of anti-Keratin 1 antibody (Poly19052) on formalin-fixed paraffin-embedded normal human skin. After antigen retrieval using sodium citrate, the tissue was incubated with the primary antibody at 1 µg/ml for 60 minutes at room temperature. BioLegend’s Ultra Streptavidin (USA) HRP Detection Kit was used for detection followed by hematoxylin counterstaining.

Description

Keratin 1 is a member of the keratin family of proteins. It is a type II cytokeratin specifically expressed in the spinous and granular layers of the epidermis along with family member Keratin 10. Mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 0.1 - 10 μg/ml is suggested. For Western blotting, a concentration range of 0.25 - 1.0 μg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
