Whole cell lysates (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel. As a control, cell lines with no/low expression (MCF-7 and SH-SY5Y) of LAT1/SLC7A5 were included. The gel was transferred to a PVDF membrane and probed with Purified anti-LAT1/SLC7A5 (clone W22208B). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG antibody (Cat. No. 405405). Direct-Blot™ HRP anti-β-Actin antibody (Cat. No. 664803) was used as a loading control.     IHC staining with anti-LAT1/SLC7A5 (clone W22208B) was performed on formalin-fixed paraffin embedded human testis tissue following antigen retrieval using Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704). The tissue sections were incubated with Purified anti-LAT1/SLC7A5 (clone W22208B) (panel B and C) followed by Alexa Fluor® 647 Goat anti-rat (Cat. No. 405416) or Rat IgG2b, κ Isotype Control (Cat. No. 400602) (panel A). Nuclei were counterstained with DAPI (Cat. No. 422801) (panel A and merged in panel C). Scale Bar: 50 µm

Cells were stained with Purified anti-LAT1/SLC7A5 (clone W22208B) (panel B and C) or Rat IgG2b, κ Isotype Control (Cat. No. 400602) (panel A) followed by Alexa Fluor® 647 Goat anti-rat (Cat. Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunohistochemistry, a concentration range of 1.25 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using either PFA-Methanol or Methanol only. We do not recommend PFA-Triton X-100 for fixation/permeabilization.

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).

For use of this antibody in intracellular Flow Cytometry (ICFC), we recommend fixation/permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401).

LAT1 is a transmembrane protein that consists of 507 amino acids and has a molecular weight of approximately 55 kDa
Distribution

LAT1 is primarily localized in the plasma membrane of cells. It is highly expressed in the blood-brain barrier and the placenta, facilitating the transport of essential amino acids to the brain and fetus. LAT1 is also found in cancer cells, where it supports rapid cell growth and metabolism
