Description

Myelin-associated glycoprotein (MAG) is a cell membrane glycoprotein and a member of the siglec (sialic acid-binding immunoglobulin-like lectins) family. MAG has two isoforms in myelinated structures in the nervous system. MAG is expressed on oligodendrocytes and Schwann cells and is believed to be involved in myelination during nerve regeneration. MAG knockout mice develop quite severe pathological abnormalities associated with myelin instability.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 5.0 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 5.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Structure
MAG is a 626 amino acid protein with a molecular mass of 70 kD.
Distribution

Tissue distribution: Central and peripheral nervous system, in periaxonal myelin and at Schmidt-Lanterman incisures.
Cellular distribution: Plasma membrane

Function
MAG is involved in the process of axon myelination and maintenance of myelin sheath along axons. MAG also functions as a receptor that promotes differentiation, maintenance and survival of oligodendrocytes. Aberrant immunoreactivity to MAG has been suggested to play a role in the pathogenesis of multiple sclerosis.
Interaction
MAG binds to neuronal sialic acid-containing gangliosides and to glycoproteins RTN4R and RTN4RL2.
