    (A) Total cell lysates from SH-SY5Y and tissue lysates from mouse and rat brain (15 µg protein each), and (B) 50 ng recombinant mouse OMG protein, mouse brain (M. brain) and rat brain (R. brain) total protein (15 µg each) were treated with or without 100 µg PNGase F at 37°C overnight to cleave the N-linked glycans from the proteins. All proteins were resolved by 4-12% Bis-tris gel electrophoresis, transferred to nitrocellulose, and probed with 1 µg/mL of purified anti-OMG antibody (clone A16003A). Proteins were visualized using a goat anti-rat-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody was used as a loading control.

Description

Oligodendrocyte myelin glycoprotein, also known as OMG, is a glycosylphosphatidylinositol (GPI) anchored protein expressed by oligodendrocytes, neurons, and astrocytes. OMG, along with Nogo (Neurite outgrowth inhibitor) and MAG (myelin-associated glycoprotein), belongs to a group of molecules located in central nervous system (CNS) myelin protein fractions with axon outgrowth inhibitory activity that prevents axonal regeneration after CNS injury. The neurite outgrowth inhibitory activities of these inhibitory molecules are mediated by binding to a common receptor complex consisting of the Nogo receptor (NgR) and the p75 neurotrophin receptor (NGFR). OMG also associates with Paired immunoglobulin-like receptor B (PirB). OMG contributes to the interactive process required for myelination in the central nervous system. OMG is also involved in the regulation of synaptic plasticity and is implicated in the inhibition of cell proliferation. The highly conserved leucine-rich repeat domain is required for inhibitory effect on both axon outgrowth and cell proliferation. Human OMG cDNA encodes a 440 amino acid residue GPI-anchored protein with a 24 amino acid signal peptide, eight leucine-rich repeats followed by five serine/threonine-rich repeats, and 23 amino acid propeptide. OMG has eleven potential N-glycosylation sites.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.2 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody also detects a non-specific protein of approximately 145 kD in the mouse brain. Clone A16003A does not cross-react with human and rat (in-house tested).
