Whole cell extracts (15 µg total protein) from the indicated cell lines/tissues were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.5 µg/mL of Purified anti-Profilin-1 (clone A18224E) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426316) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining of Purified anti-Profilin-1 (clone A18224E) on formalin-fixed paraffin-embedded human tonsil. Following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0), the tissue was incubated without (panel A) and with (panel B) 10 μg/mL of Purified anti-Profilin-1 (clone A18224E) followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40X objective. Scale bar: 50 μm

Profilin-1 is an actin-binding protein responsible for the regulation of actin dynamics. Profilin-1 assists in the exchange of ADP for ATP on G-actin, facilitating the polymerization of actin monomers into filaments by promoting assembly at the growing end. Thus, profilin-1 is implicated in processes like cell motility, cytokinesis, and endocytosis, highlighting its significance in maintaining cellular structure and function. Profilin-1 also interacts with numerous proteins involved in signaling pathways, influencing cellular responses to intracellular and extracellular cues. Emerging evidence also shows that profilin-1 can bind to microtubules, facilitating interactions between the two major components of the cytoskeleton. Mutations in profilin-1 have been associated with amyotrophic lateral sclerosis (ALS) and is suspected to cause insoluble aggregates due to disruption in axonal outgrowth, cell integrity and protein distribution.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody (clone A18224E) is not suitable for immunocytochemistry (ICC).
For immunohistochemistry (IHC), we recommend antigen retrieval with Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0). Antigen retrieval with Sodium Citrate is not recommended as it results in mis-localized signal. - RRID
