Total cell lysate from HeLa (15 µg/lane) were resolved by electrophoresis (4-20% Tris-Glycine), transferred to nitrocellulose, and probed with purified anti-GAPDH (clone Poly6314, rabbit IgG) antibody (lane 1) and purified anti-β-actin (clone 2F1-1, mouse IgG) antibody (lane 2). After a 30 minute wash, both lanes were further probed with purified anti-rabbit IgG (clone 6B9G9) antibody. Proteins were visualized using an HRP goat anti-mouse IgG antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin (clone 2F1-1) antibody was used as a loading control.

Immunofluorescence of HeLa cells without (A) or with (B-D) pericentrin Rabbit primary antibody (Cat. No. 923701). Rabbit IgG mouse antibody was used as the secondary antibody (Cat. No. 410404). Alexa Fluor® 488 (Green) Goat anti-Mouse IgG (Cat. No. 405319) was used as tertiary antibody. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective. Exposure time (Seconds) for (A-D) is 1/15. Concentrations for (A, B) is 1 µg/ml, (C) is 0.5 µg/ml, (D) is 0.2 µg/ml.
Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 0.5 µg per ml. For immunocytochemistry, a concentration range of 0.2 - 1.0 μg/ml (Dilution 1:500-1:2500) is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
IgG antibodies are large molecules of about 150 kD made of four peptide chains. It contains two identical class γ heavy chains of about 50 kD and two identical light chains of about 25 kD.
