Total cell lysates (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-SH2B3/LNK Antibody, clone O92B11, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

SH2B3, originally described as LNK, is a member of the SH2B adaptor family of proteins that controls signaling activities from activated growth factor and cytokine receptors. This protein functions as a negative regulator of cytokine signaling and plays a critical role in hematopoiesis and proliferation. Through an SH2 domain, SH2B3 has been shown to directly bind and inhibit downstream cytokine effectors, including Janus Kinases and other tyrosine kinases. SH2B3 functions as potent tumor suppressor in myeloproliferative disorders, while elevated SH2B3 has been described to confer a pro-survival advantage against the pro-apoptotic effects of interferon in cutaneous melanomas.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
