Capture ELISA of purified anti- Testosterone antibody (clone 3T16) and isotype-matched control mouse IgG1 binding to OVA-conjugated Testosterone (OVA-Testosterone). ELISA was performed by coating wells with 100 ng of clone 3T16. The wells were then incubated at 37°C for 1 hour with serially diluted (0.002 to 2 µg/ml) OVA-Testosterone, followed by incubation with rabbit anti-OVA antibody, and horseradish peroxidase labeled goat anti-Rabbit secondary antibody. TMB (3, 3', 5, 5' tetramethylbenzidine, Cat. No. 421501) was used as the detection system.

ELISA was performed by coating wells with 600 ng OVA-Testosterone. The wells were then incubated with clone 3T16 or mouse IgG at 37°C for 45 minutes, followed by incubation with horseradish peroxidase labeled goat anti-mouse secondary antibody. TMB (3, 3', 5, 5' tetramethylbenzidine, Cat. No. 421501) was used as the detection system.

Description

In vertebrate, Testosterone is the major male sex hormone that is essential to the development of reproductive organs and secondary sexual characteristics. It also plays a key role in muscle development, body mass increase and body fair growth. Importantly, it is critical to sperm development.

In vivo, testosterone is primarily secreted by testicles (in male), and by ovaries (in female), and eventually metabolized in liver. In the central nervous system, Testosterone is one of the key components of the hypothalamic-pituitary-adrenal axis, which is activated acutely by dominance challenger. It has been reported that different level of sex hormones, including testosterone, determines human behavior, as well as brain development and recovery after injury.

Recommended Usage

Each lot of this antibody is quality control tested by ELISA assay. For ELISA Capture applications, a concentration range of 10 - 150 ng/mL is recommended. For Direct ELISA applications, a concentration range of 1.0 - 10 ng/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

This antibody has been tested against human and mouse sera.
