IHC staining of Alexa Fluor® 647 anti-α Smooth Muscle Actin (clone 1A4) on formalin-fixed paraffin-embedded human kidney tissue. Following antigen retrieval using 1X Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704), the tissue was incubated with Alexa Fluor® 647 anti-α Smooth Muscle Actin (clone 1A4) overnight at 4°C. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 10X (panel B) or 40X (panel A) objective. Scale bar: 50 µm

    NIH/3T3 cells were treated with (panels A and B) or without 5 ng/mL of Recombinant Mouse TGF-β1 (Cat. No. 763104) (panels C and D) for 48 hours at 37°C, fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes, permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Alexa Fluor® 647 anti-α Smooth Muscle Actin (clone 1A4) (panels A and C). Nuclei were counterstained with DAPI (Cat. No. 422801). Images were merged (panels B and D). The images were captured by a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm

Description

Cytoplasmic actins, which belong to the microfilament system of cytoskeleton proteins, are some of the most conserved eukaryotic proteins expressed in mammals. α-smooth muscle actin is widely expressed in contractile cells, most notably the mural cells that line arteries and arterioles. Overexpression of α-smooth muscle actin is a hallmark of fibrosis, a thickening or scarring of damaged tissue.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of  5.0 - 10.0 µg/mL is suggested.  For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.

Application Notes

For use in immunohistochemistry (IHC) on formalin-fixed paraffin-embedded tissues, we recommend heat-induced antigen retrieval with either Tris-EDTA pH 9.0 or Citrate Buffer, 10X (Cat. No. 420902).

For use in immunocytochemistry (ICC), we recommend fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with 0.5% Triton-X.
