Osteocalcin is the bone’s most abundant non-collagenous protein. It is produced by osteoblasts as a 49 amino acid protein with 3 glutamine acid residues (Glu 17, 21, and 24) that are gamma carboxylated in a vitamin K-dependent manner. The fully carboxylated protein has a high affinity for the bone extracellular matrix. Bone resorption results in a decrease in pH, causing the decarboxylation of osteocalcin. This generates partially decarboxylated (undercarboxylated), or fully decarboxylated (uncarboxylated) osteocalcin. These forms of osteocalcin have decreased affinity for the bone extracellular matrix and are released into the bloodstream, allowing it to bind the receptor GPCR6a, acting as hormones promoting insulin sensitivity, glucose tolerance, and testosterone biosynthesis. In patients with diabetes, reduced serum levels of osteocalcin are negatively correlated with obesity and insulin resistance.
BioLegend’s ELISA MAX™ Deluxe Set is a sandwich Enzyme-Linked Immunosorbent Assay (ELISA). A human uncarboxylated osteocalcin specific monoclonal antibody is first coated on a 96-well plate. Standards and samples are added to the wells, and uncarboxylated osteocalcin binds to the immobilized capture antibody. Next, a biotinylated anti-human osteocalcin (uncarboxylated) detection antibody is added, producing an antibody-antigen-antibody “sandwich”. Avidin-horseradish peroxidase is subsequently added, followed by TMB Substrate Solution, producing a blue color in proportion to the concentration of uncarboxylated osteocalcin present in the sample. Finally, the Stop Solution changes the reaction color from blue to yellow, and the microwell absorbance is read at 450 nm with a microplate reader.
