In mammals, proteins of the 14-3-3 family include isoforms β, γ, ε, σ, ζ, τ, and η. 14-3-3 δ isoform is the phosphorylated form of 14-3-3 ζ. These highly conserved proteins form homo- or heterodimers with many transcription factors, metabolic enzymes, kinases, phosphatases, and other signaling molecules, generally through a phospho-Ser/Thr motif. The binding regulates the location, stability, interaction with other molecules, and phosphorylation status of the target proteins which suggests the important roles for these 14-3-3 family proteins on regulation of cellular functions and responses. Growing data demonstrate that the specific temporal and spatial distributions of 14-3-3 family proteins are critical for developmental processes; and in some cancers, the regulation of these proteins are different from healthy controls. 14-3-3 ζ interacts with IRS1 protein and is involved in the PI3K-Akt signaling pathway.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 – 2.0 µg/ml in human, mouse, and rat and 0.25 – 2.0 µg/ml in Drosophila. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/ml is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

14-3-3 family members share a high homology in amino acid sequences. Hence, this clone may recognize other 14-3-3 family members with predicted MW of 27, 28kD.
