Whole cell extracts (15 µg protein) from untreated (-) or LY294002-treated (+, 100 µM for 1 hour) HEK-293 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-4EBP1 Phospho (Thr70) Antibody, clone A18010A, for 2 hours at room temperature. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

In eukaryotes, the mTOR kinase signaling pathway links cell growth and proliferation with nutrient availability. Under nutrient-deplete conditions, eukaryotic translation initiations factor 4E-binding protein 1 (4EBP1) binds the general translation factor eIF4E, resulting in an attenuation of cap-dependent translation. Under nutrient-replete conditions, mTORC1 and mTORC2 complexes phosphorylate 4EBP1 at Threonine 70 and disrupt the formation of the 4EBP1-eIF4E complex, thereby alleviating the repressive function of 4EBP1.
