Protein extracts (15 µg total protein) from the indicated cell lines and tissues were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL of purified anti-Aldolase C (clone A18070A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining of purified anti-Aldolase C (clone A18070A) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using 1X Tris-EDTA, pH 9.0, the tissue was incubated without (panel A) and with (panel B) 1 µg/mL of purified anti-Aldolase C (clone A18070A) overnight in 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 20X objective. Scale bar: 50 µm -
Cells were fixed with 4% PFA Fixation Buffer (Cat. No. 420801) and permeabilized with methanol for 10 minutes, and then blocked with 5% FBS for 1 hour at room temperature. A549 cells (low expression control, panel B and D) and U87-MG cells (positive control, panel A and C) were then stained with 5.0 µg/mL of purified mouse IgG2b, κ isotype control (Cat. No. 402202) (panels A and B) or purified anti-Aldolase C (clone A18070A), followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 60X objective. Scale bar: 50 µm

Aldolase C is one of the three aldolase isozymes and plays a key role in ATP biosynthesis. It participates in glycolysis and gluconeogenesis by catalyzing the conversion of fructose-1-6-biphosphate to glyceraldehyde-3-phosphate, or glyceraldehyde, and di-hydroxy-acetone-phosphate. In addition, it is part of the stress-response pathway induced by hypoxia. It has been found that Aldolase C is upregulated in the brains of schizophrenia patients. Moreover, inactivation of Aldolase C by oxidation has been reported in mild cognitive impairment (MCI) and Alzheimer’s disease (AD).

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 5.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 2.5 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Antigen Details
- Structure
- Aldolase C is a 364 amino acid protein with an expected molecular weight of 39 kD.It is a member of the three aldolase isozymes. Aldose C shares 68% identity with Aldolase B and 78% with Aldolase A. The catalytic site is highly conserved in all three isoforms.
- Distribution
-
Although Aldolase C is widely expressed, it is predominantly expressed in the hippocampus, Purkinje cells, neurons of the brain, and smooth muscle.
- Function
- Glycolytic enzyme that catalyzes the reversible aldol cleavage of fructose-1,6-biphosphate and fructose 1-phosphate to dihydroxyacetone phosphate and either glyceraldehyde-3-phosphate or glyceraldehyde, respectively.
