Whole cell extracts (15 µg protein) from HeLa cells transfected with non-targeting control siRNA (siCON) or siRNA targeting Arginase 1 (siARG1) were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 0.5 µg/mL (1:1000 dilution) of Purified anti-Arginase 1 Antibody, clone O94E6/ARG1, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-Actin Antibody (Cat. Proteins were visualized using chemiluminescence detection by incubation with HRP Goat anti-mouse secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-GAPDH was used as a loading control (Cat. No. 649203, 1:1000 dilution).

    IHC staining using purified anti-Arginase 1 antibody (clone O94E6/ARG1) on formalin-fixed paraffin-embedded human liver tissue. The tissue was incubated with 10 μg/mL of anti-Arginase 1 antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG antibody (red) (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40X objective. Scalebar = 50 μM.

Description

ARG1 (Arginase 1), is a marker for the M2 anti-inflammatory macrophage subset that hydrolyzes l-arginine into urea and ornithine, and plays an important role in wound healing, antiviral immune responses, and atherosclerosis. The inherited deficiency of this enzyme results in argininemia, which is an autosomal recessive disorder characterized by hyperammonemia. Recent studies show that intracellular pathogens induce the expression of ARG1 in mouse macrophages through the Toll-like receptor (TLR) pathway. The deletion of this enzyme in macrophages favors host survival. Other studies indicate that ARG1 also plays a role in T cell responses and selective antagonists of ARG1 might prove beneficial in fighting diseases in which T-cell responses are inappropriately suppressed.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per mL. For immunohistochemistry, a concentration range of 1 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
